چکیده

Nuclei of hemlock (Tsuga canadensis and Tsuga canadensis var. nana) were investigated for the presence of flavanols. Histochemical staining with pdimethylaminocinnamaldehyde proved to be a highly valuable method yielding a bright blue flavanol coloration for nuclei. There was a significant variation in flavanol deposition (1) among nuclei, (2) at the subnuclear level and also (3) along the chromosomes during mitosis. The presence of flavanols in nucleoli could not be established probably because they were too small, measuring less than 1 μm in diameter. In contrast to Tsuga, the cells and nuclei of rootlets from rye (Secale cereale) were totally devoid of natural flavanols. However, externally added flavanols, catechin and epicatechin, were bound to the rye nuclei, while the rather large nucleoli failed to associate with the flavanols. The strong sink activity of nucleoplasm and chromosomes for flavanols in Tsuga and Secale indicates a process which is apparently widespread even in distantly related plant species. Variations in chromatin-associated flavanols could to some extent be induced by acetylation/deacetylation of histones, as confirmed in the present study by means of UVVIS spectroscopic titrations of histone sulphate and chemically acetylated histone sulphate.

Nuclei of Tsuga canadensis: Role of Flavanols in Chromatin Organization

Needle primordia of Tsuga canadensis (hemlock) arising from flank meristems of a shoot apex, form cell lineages consisting of four or eight cells. Within a recently established lineage there is striking uniformity in the pattern of nuclear flavanols. This fact points to an identical transcriptional expression of these flavanols during cell cycling. However two lineages, even if located close to...

Chromosomal Variation in Three Human-Mouse Hybridoma Cell Lines after Various Passaging Intervals as Assessed with Two Different Staining Methods

Objective(s) The main objective of this study was to investigate the status of chromosome stability in 3 human-mouse hybridoma cell lines over a period of time in various passages. Materials and Methods Metaphase spreads from 3 human-mouse cell lines (HF2X653, SPMO-4 and F3B6) that had been cultured in 4 successive passages, from 1 to 4 weeks, were prepared and analyzed. Metaphase chromosome...

Tissue inhomogeneity in proton therapy and investigation of its effects on BRAGG peak by using MCNPX code

Background: Hadron therapy for malignant tumor is becoming increasingly popular. There are many factors which effect on implementation of a proper treatment planning. The purpose of this work is to investigate the inhomogeneity effects as affecting factor on proton range, Full width at half maximum (FWHM) and 20% position of penumbra (P20) by MCNPX code. Materials and Methods: An inhomogeneous ...

Estimation of seed viability constants for tall wheatgrass, cocksfoot, rye, and sheep fescue to inform gene banking decisions. Hamid Reza Eisvand

Stored seeds deteriorate over time and must be regenerated to ensure that the benefits of ex situ conservation are realized. Prediction of seed longevity is based on the seed viability equation. This equation has four constants which are species specific. The aim of this project is the estimation of these constants and prediction of regeneration frequency for Elytrigia elongata, Dactylis glomer...

Identification and application of anthocyanine from wild safflower (Carthamus oxyacanthus) seed coat color

Background & Aim: Despite valuable medicinal properties of safflower, breeding new varieties and development of the crop culture area has been limited due to the lack of genetic variation for some crucial characteristics as resistance to diseases and pests like safflower fly. In this study, populations of wild safflower, C. oxyacanthus were collected from western, central and ...
